چکیده

This study aims to investigate the role of long non-coding RNA (lncRNA) HOX transcript antisense (HOTAIR) in regulation anoikis resistance ovarian cancer cells, a prerequisite for metastasis and chemoresistance cells. Ovarian SKOV3 cells were cultured an ultra-low attachment system establish model. The relationship between cellular capability HOTAIR expression level was studied by flow cytometry RT-PCR. ability spheroid formation, migration, invasion suspended assessed following knockdown expression. EZH2, H3K27me3, representative targets anoikis-related biomarkers also detected. An increase duration suspension culture time rendered anoikis-resistant with significantly lower apoptotic rate compared adherent increased significantly, while that did not. Following small interfering (siRNA)-mediated expression, abilities resistance, decreased Knockdown levels reduced forming tumor continuous cultures. Moreover, EZH2 correlated thus regulating miR-193a DOK2 via introducing H3K27me3. Western blot analysis markers showed N-cadherin, ZEB1, TWIST1 downregulated inhibition HOTAIR, E-cadherin ErbB3 upregulated. In conclusion, enhances recruiting influencing H3K27 methylation may contribute invasion,

EZH2 Methyltransferase and H3K27 Methylation in Breast Cancer

Histone modifications are thought to control the regulation of genetic programs in normal physiology and cancer. Methylation (mono-, di-, and tri-methylation) on histone H3 lysine (K) 27 induces transcriptional repression, and thereby participates in controlling gene expression patterns. Enhancer of zeste (EZH) 2, a methyltransferase and component of the polycomb repressive complex 2 (PRC2), pl...

Functions of lncRNA HOTAIR in lung cancer

Long non-coding RNAs (lncRNAs) govern fundamental biochemical and cellular processes. lncRNA HOX transcript antisense RNA (HOTAIR) represses gene expression through recruitment of chromatin modifiers. The expression of HOTAIR is elevated in lung cancer and correlates with metastasis and poor prognosis. Moreover, HOTAIR promotes proliferation, survival, invasion, metastasis, and drug resistance ...

LncRNA HOTAIR controls the expression of Rab22a by sponging miR-373 in ovarian cancer

Increasing evidence suggests that the long non-coding RNA, HOX transcript antisense intergenic RNA (HOTAIR) is widely involved in the progression and metastasis of cancer. However, the specific role of HOTAIR in ovarian carcinogenesis remains to be fully elucidated. In the present study, the levels of HOTAIR were detected in 30 paired cancer and noncancer tissues using reverse transcription-qua...

Genetic variants of lncRNA HOTAIR and risk of epithelial ovarian cancer among Chinese women

Ovarian cancer is one of the common female malignant tumors globally. However, exactly mechanism of ovarian cancer remained unknown. HOTAIR, a lncRNA in the mammalian HOXC locus, has been fully explored for its genetic variants, expression level and carcinogenesis, development and progression of multiple cancers, except for ovarian cancer. In this study, we hypothesized that abnormal expression...

LncRNA GAS5 suppresses ovarian cancer by inducing inflammasome formation

OBJECTIVE Long non-coding RNA growth arrest-specific transcript 5 (lncRNA GAS5) is involved in various kinds of cancer. However, the role of lncGAS5 in the development of ovarian cancer remains unclear. In the current study, we explored the cellular mechanism and clinical value of lncRNA GAS5 in ovarian cancer. METHODS Quantitative real-time PCR was used to detect mRNA level of LncRNA GAS5 in...
